پژوهشگران دانشگاه پادووا، دانشگاه ونیز ایتالیا، دانشگاه هاروارد، و دانشگاه کلورادو ایالات متحده امریکا در پژوهشی مشترک به بررسی وراثت‌پذیری تاب‌آوری مغز انسان در برابر آسیب اختلالات در پژوهش های دوقلوها در انسان های سالم پرداختند.

روش پژوهش وراثت‌پذیری تاب‌آوری مغز انسان:

  • در این پژوهش پیمایش تصویر برداری 1206 فرد سالم شرکت نمودند. هسته پژوهش شامل دو گروه دو قلوی سالم با جمعیت 453 و 463 نفر  بودند.
  • برای تهیه کانکتوم (دیاگرام اتصالات) مغزی کارکردی از fMRI حالت استراحت استفاده شد. شدت میدان مغناطیسی MRI به اندازه 3 تسلا بود.
  • همچنین نقشه مغزی از سطح کرتکس، ضخامت کرتکس، انحنای کرتکس، عمق شکنج ها، و میزان میلینه شدن آکسون ها تهیه شد.
  • برای تهیه نقشه های مغزی، از 32 هزار برش و 180 ناحیه مغزی در هر نیمکره استفاده شد.

یافته های پژوهش وراثت‌پذیری تاب‌آوری مغز انسان:

  1. برای چندین شاخص نقشه مغزی کانکتوم، شاخص های وراثت پذیری متوسط بدست آمدند.
  2. درزمینه وراثت‌پذیری تاب‌آوری مغز انسان، شاخص های به دست آمده بر اساس حذف اتصالات مغزی بودند، نه بررسی نواحی خاص مغز.
  3. تاثیرات ژنتیک بر وراثت‌پذیری تاب‌آوری مغز انسان بیش از تاثیرات محیطی اکتسابی است.
  4. میزان وراثت‌پذیری تاب‌آوری مغز انسان به صورت ژنتیک برای شبکه حالت پایه، شبکه بینایی، و مدارهای حسی-حرکتی مغز بالا و معنادار است.

راهبردهای کارکردی پژوهش وراثت‌پذیری تاب‌آوری مغز انسان:

  • به نظر می رسد وراثت‌پذیری تاب‌آوری مغز انسان نشانگر دلایل اسیب پذیری و حساسیت متفاوت افراد نسبت به بیماری های مغزی و اختلالات روانپزشکی است.
  • طبق نقشه مغزی از نظر ژنتیک مغزی افراد در برابر بیماری های نورولوژیک، انواع آسیب های مغزی، و بیماری های روانپزشکی توان مقاومت متفاوتی دارد که به نوعی بیانگر تفاوت های فردی در نقشه مغزی هستند.
  • تحقیقات گسترده تری برای ایجاد خط پایه در نقشه مغزی انسان و روش های سنجش ساده تر و ارزان تر آن لازم است تا بتوان در سطح وسیع جمعیتی میزان مقاومت و تاب آوری علیه آسیب های مغزی را اندازه گیری نمود.
  • در صورتی که میزان تاثیر عوامل ژنتیک بر وراثت‌پذیری تاب‌آوری مغز انسان بالا باشد، لازم است عوامل حمایتی برای گروه های در معرض خطر به شکلی دقیق تر در سطح اجتماعی و خدمات بهداشتی فراهم شود تا میزان اختلالات مغزی، عصبی و روانی در جامعه کاهش یابند.

Heritability of brain resilience to perturbation in humans

Highlights

  • Strokes, traumatic lesions, tumors and neurodegenerative disorders are all examples of neurological conditions that can severely impair the normal functioning of the human brain.
  • Given the same amount of damage, a lot of variability exists in individual outcomes, a phenomenon often attributed to so-called “brain resilience”.
  • By simulating brain lesions in a large dataset of twins, we investigated whether brain resilience is mainly influenced by genetic factor or by environmental factors.
  • Results imply both heritable and environmental components of brain resilience, which may be important for designing interventions to help build individual resilience.

Abstract

Objective:

Resilience is the capacity of complex systems to persist in the face of external perturbations and retain their functional properties and performance.

Aim:

In the present study, we investigated how individual variations in brain resilience, which might influence response to stress, aging and disease.

They are influenced by genetics and/or the environment, with potential implications for the implementation of resilience-boosting interventions.

Method:

Resilience estimates were derived from in silico lesioning of either brain regions or functional connections constituting the connectome of healthy individuals belonging to two different large and unique datasets of twins, specifically:

463 individual twins from the Human Connectome Project and 453 individual twins from the Colorado Longitudinal Twin Study.

Results:

As has been reported previously, moderate heritability was found for several topological indexes of brain efficiency and modularity.

Importantly, evidence of heritability was found for resilience measures based on removal of brain connections rather than specific single regions.

It is suggesting that genetic influences on resilience are preferentially directed toward region-to-region communication rather than local brain activity.

Specifically, the strongest genetic influence was observed for moderately weak, long-range connections between a specific subset of functional brain networks:

  • the Default Mode
  • Visual networks
  • Sensorimotor networks

Conclusion

These findings may help identify a link between brain resilience and network-level alterations observed in neurological and psychiatric diseases, as well as inform future studies investigating brain shielding interventions against physiological and pathological perturbations.
