پژوهشگران دانشگاه یونسِی و دانشگاه زنان ایوها کرده جنوبی در پژوهشی که به تازگی منتشر شده است به بررسی تاثیرات متقابل تمرینات ورزشی و سالمندی در حوزه کاهش پیری و تقویت سیستم ایمنی پرداختند.

??? روش پژوهش

استرس اکسیداتیو عامل اصلی تخریب سلول‌ها، بافت‌های بدنی، و در نهایت مرگ موجود زنده است. نتیجه بسیار خطرناک استرس اکسیداتیو تخریب تدریجی سیستم ایمنی بدن است.

پژوهش آزمایشی حاضر بر مدل موش صحرایی صورت گرفت تا به بررسی عوامل موثر در کاهش استرس اکسیداتیو و کاهش تخریب سیستم ایمنی بپردازد. شاخص پیری توسط AGEs و شاخص فعالیت سیستم ایمنی بر اساس فنوتیپ سلول‌های T تیموس و طحال نحاسبه شد.

???یافته های پژوهش

  1. تمرینات ورزشی و سالمندی با یکدیگر رابطه عکس دارند. هرچه سطح تمرینات ورزشی روزانه بالاتر رود، روند سالمندی کندتر می شود.
  2. پس از ۶ هفته مصرف مداوم d-galactose سطح شاخص‌های پیری در نمونه‌های مورد آزمایش به شکل معناداری افزایش یافت.
  3. پس از ۶ هفته مصرف مداوم d-galactose سطح فعالیت سیستم ایمنی در طحال کاهش معناداری یافت.
  4. پس از ۴ هفته ورزش و مصرف ویتامین E، سطوح سلولی و خونی استرس اکسیداتیو به میزان بسیاری کاهش یافتند.
  5. مهمتر از همه، در گروهی که تنها ورزش داشتند، سطح تخریب فعالیت سیستم ایمنی به میزان زیادی کاهش یافته و فنوتیپ سلول‌های T بهبود یافتند.

 

?????? راهبردهای کارکردی:

  • ورزش داوطلبانه روزانه، به تنهایی می‌تواند تاثیر منفی استرس اکسیداتیو بر سیستم ایمنی را خنثی نماید و در حقیقت ورزش‌ روزانه بهترین داروی ضد بیماری و ضد پیری است.
  • لازم است در سبک زندگی جامعه، ورزش منظم روزانه وارد شود و مشوق‌ها و سیاستگذاری‌های لازم برای تشویق اجتماعی ورزش روزانه تنظیم شوند تا به این ترتیب پیری و بیماری‌ها و هزینه‌های سربار وابسته به آن در سطح جامعه محدود شود.

 

Voluntary exercise reverses immune aging induced by oxidative stress in aging mice

Abstract

Excessive oxidative stress leads to aging due to persistent damage to the cells, tissues, and the entire organism. Immunosenescence is also a devastating consequence of oxidative stress, but there is a lack of research on effective ways to overcome it.

In this study, we used physiologic and immunological aging mouse models that had sustained oxidative stress to investigate whether voluntary exercise and/or antioxidant treatment could overcome oxidative damage as well as aging. We established an aging model induced by continuously administering d-galactose (d-gal) to 6-week-old female C57BL/6J mice.

We also assessed reversal of immunosenescence by providing free-wheel running and vitamin E (vit E) supplementation to this aging model. As an aging index, the level of advanced glycation end products (AGEs) in the blood was measured. Phenotypes of T cells in the thymus and spleen were examined as an index of immunosenescence.

Intracellular reactive oxygen species (ROS) levels in the mouse spleen and levels of AGEs in the blood were significantly higher after 6 weeks of d-gal administration. In addition, immunosenescence was observed, in which the naïve:effector cell ratio in the spleen decreased. After 4 weeks of free-wheel running and vit E administration, both intracellular ROS and serum AGE levels decreased.

Above all, free-wheel running restored the naïve:effector ratio of cytotoxic T lymphocytes reduced by d-gal administration. Taken together, these results suggest that voluntary exercise may be effective in restoring immunosenescence induced by oxidative stress.
